2、蛋白C末端49個(gè)氨基酸,即NP49)多肽作為包被抗原,建立標(biāo)記疫苗免疫豬血清中NP49抗體的ELISA鑒別診斷方法。通過對(duì)NP49一ELISA工作條件的優(yōu)化,結(jié)果顯示NP49一ELISA的多肽抗原最適包被濃度為500ng/孔,被檢血清最佳稀釋度為1:40。利用ROC曲線法確定S/P臨界值為0.2,該方法批內(nèi)與批間重復(fù)試驗(yàn)的變異系數(shù)均小于10%,與幾種常見豬病陽性血清無交叉反應(yīng),具有較好重復(fù)性和特異性。本研究建立的NP49.ELISA鑒別診斷方法為豬繁殖與呼吸綜合征新型基因標(biāo)記疫苗的臨床應(yīng)用提供了有力保障。關(guān)鍵詞:豬繁殖與呼吸綜合征標(biāo)記疫苗;多肽抗原;NP49一ELISA;鑒別診斷中圖分
