1、·438·工程雜志2013年l2月第19卷第期h』旦i塑Eg:旦b叢2Q1!:!Q:論著小干擾RNA靶向沉默CXCR2基因抑制肝癌細胞的增殖謝斌輝王小農(nóng)劉鳳恩何曉謝元康周奇【摘要】目的研究CXCR2一小干擾RNA(siRNA)基因體外抑制肝癌細胞增殖的作用。方法將CXCR2.siRNA以脂質(zhì)體轉(zhuǎn)染法轉(zhuǎn)染肝癌細胞hepG:,24h后熒光顯微鏡觀察細胞熒光含量。采用RT—PCR及免疫印跡法分別檢測正常肝Chang細胞、未轉(zhuǎn)染肝癌hepGz細胞以及轉(zhuǎn)染CXCR2一siRNA基因后hepG2細胞在轉(zhuǎn)染24、48、72hA的CXCR2mRNA和蛋白表達水平。以未轉(zhuǎn)染的hepG2細胞為對照.CCK8
2、法檢測轉(zhuǎn)染CXCR2。siRNA的hepG2細胞在轉(zhuǎn)染24、48、72h后的增殖。以未轉(zhuǎn)染的hepG:細胞為對照,檢測轉(zhuǎn)染CXCR2一siRNA24h后hepGz細胞與Transwell小室孵育24h時的侵襲能力。結(jié)果CXCR2一siRNA轉(zhuǎn)染hepG224h后鏡下可見大量綠色熒光,轉(zhuǎn)染率為80%。hepG2細胞CXCR2mRNA和蛋白表達水平高于Chang細胞(mRNA:1.69_+0.22比0.63_+0_31,蛋白:1.93_+0.25比0.84_+0.29,均P<0.05)。轉(zhuǎn)染CXCR2.siRNA24和48h后hepG2細胞CXCR2mRNA表達量低于未轉(zhuǎn)染的hepG2細胞(
3、0.75±0.24、0.83±0.21比1.78_+0.25,均P<0.05),72h后CXCR2一siRNA雖然仍能抑制hepG2細胞CXCR2mRNA的表達,但與未轉(zhuǎn)染的hepG2細胞相比差異并無統(tǒng)計學意義(1.35_+0.18比1.78_+0.25,P>0.05)。轉(zhuǎn)染CXCR2一siRNA24和48h后的hepG2細胞CXCR2蛋白表達低于未轉(zhuǎn)染的hepG2細胞(0.91_+0.25、1.16_+0.23比1.98_+0.31,均P0.05)。轉(zhuǎn)染CXCR2一s
4、iRNA24、48、72h后hepG2細胞mRNA和蛋白表達差異均無統(tǒng)計學意義。與未轉(zhuǎn)染的hepG2細胞相比,轉(zhuǎn)染CXCR2一siRNA24、48、72h后hepG2細胞增殖受到抑制(均P<0.05)。轉(zhuǎn)染CXCR2一siRNA的hepG2細胞對Transwell小室的侵襲能力明顯低于未轉(zhuǎn)染的hepG:細胞[(36_+5)個/高倍視野(x200)比(526_+9)個/高倍視野(×200),P<0.05]。結(jié)論siRNA沉默CXCR2基因體外可有效抑制肝癌細胞的增殖?!娟P(guān)鍵詞】RNA,小分子干擾;受體,白細胞介素8B;肝腫瘤,實驗性;基因沉默TargetedsilencingofCXCR2
