1、.640.安徽醫(yī)藥AnhuiMedicalandPharmaceuticalJournal2015Apr;19(4)沒(méi)藥甾酮對(duì)膽管癌RBE細(xì)胞移植瘤生長(zhǎng)和miRNA表達(dá)譜的影響仲飛,童鑄廷,范璐璐,姚夢(mèng)群,孫國(guó)平(安徽醫(yī)科大學(xué)第一附屬醫(yī)院,安徽合肥230022)摘要:目的研究沒(méi)藥甾酮對(duì)膽管癌RBE細(xì)胞株移植瘤生長(zhǎng)和miRNA表達(dá)的影響。方法雌性SD大鼠建立膽管癌RBE細(xì)胞株移植瘤模型,隨機(jī)分為對(duì)照組和沒(méi)藥甾酮高、低劑量實(shí)驗(yàn)組;高、低劑量實(shí)驗(yàn)組大鼠分別予40、20mg·kg~·d沒(méi)藥甾酮灌胃,對(duì)照組
2、大鼠予生理鹽水灌胃。每5天測(cè)量裸鼠質(zhì)量和腫瘤體積,處理30d后取材,提取腫瘤組織總RNA并分離miR—NA。采用微陣列雜交,通過(guò)芯片掃描和數(shù)據(jù)分析獲得miRNAs表達(dá)譜。結(jié)果高、低劑量實(shí)驗(yàn)組大鼠移植瘤體積分別在處理20d和25d后顯著低于對(duì)照組(P<0.01);處理30d后高、低劑量組移植瘤質(zhì)量均顯著低于對(duì)照組(P<0.01)。芯片結(jié)果顯示,高劑量實(shí)驗(yàn)組瘤組織miRNAs表達(dá)譜與對(duì)照組相比有顯著差異。結(jié)論沒(méi)藥甾酮可以顯著抑制膽管癌RBE細(xì)胞株移植瘤生長(zhǎng)并影響其miRNA的表達(dá)。關(guān)鍵詞:沒(méi)藥甾酮;膽
