2、技術(shù)從銀杏葉片中分離到黃酮3’羥化酶基因GbF3’H的cDNA全長(zhǎng)序列(GenBankNo.:KP056747)。其全長(zhǎng)為2144bp,含有一個(gè)1671bp的開放式閱讀框(ORF),編碼556個(gè)氨基酸序列,預(yù)測(cè)氨基酸大小為63.47kD,等電點(diǎn)為7.71。蛋白質(zhì)同源序列分析表明,GbF3’H與其他物種F3’H同源性較低,而與菊苣(Cichoriumintybus)同源性最高,為56.3%。同源建模分析顯示GbF3’H與P450家族蛋白的三維結(jié)構(gòu)及活性位點(diǎn)高度相似,最終定位于微粒體膜上。進(jìn)化樹分析結(jié)果顯示GbF3’H與其他植物分
3、化較早,序列差異較大。組織表達(dá)分析顯示,GbF3’H在銀杏不同組織中都有表達(dá),其中雄蕊表達(dá)水平最高,其次為成熟葉。誘導(dǎo)表達(dá)分析顯示,GbF3’H轉(zhuǎn)錄水平受UV-B、6-BA、SA、ABA和IAA影響誘導(dǎo)上調(diào),而傷害處理對(duì)其表達(dá)量無(wú)明顯影響。GbF3’H誘導(dǎo)表達(dá)模式與調(diào)控銀杏黃酮含量變化呈正相關(guān),其上游調(diào)控序列存在與黃酮調(diào)控相關(guān)的順式元件,意味著GbF3’H可能參與了銀杏黃酮類物質(zhì)的合成代謝,并與黃酮類物質(zhì)向花青素合成分流有關(guān)。關(guān)鍵詞:銀杏;類黃酮3’羥化酶;黃酮;表達(dá)調(diào)控中圖分類號(hào):S664.3文獻(xiàn)標(biāo)志碼:A文章編號(hào):0513
