2、A+-11mg·L6-BA的培養(yǎng)基上,經(jīng)過180d的培養(yǎng),誘導(dǎo)出3種愈傷組織:Ⅰ,淺黃色松散易碎;Ⅱ,白色疏松半透明水漬狀;Ⅲ,黃色致密顆粒狀。對愈傷組織繼代培養(yǎng)基中的2,4-D、NAA和6-BA濃度進(jìn)行優(yōu)-1-1-1化,結(jié)果表明培養(yǎng)基中含有1mg·L2,4-D、0.25mg·LNAA、0.25mg·L6-BA時可獲得胚性狀態(tài)良-1-1好的愈傷組織。胚性愈傷組織在MS無機(jī)鹽+0.25mg·LNAA+0.5mg·LTDZ+維生素B5+100mg·-1-1-1-1L谷氨酰胺+230mg·L脯氨酸+100
3、mg·L麥芽提取物+45g·L蔗糖的體胚誘導(dǎo)培養(yǎng)基中培養(yǎng)20d后,轉(zhuǎn)移到MS基本培養(yǎng)基上培養(yǎng)30d,1g胚性愈傷組織可獲得50~60個體胚。成熟體胚在MS+0.2-1-1mg·LIAA+0.5mg·L6-BA的萌發(fā)培養(yǎng)基上培養(yǎng)20d時,體胚萌發(fā)率為85%。萌發(fā)的體胚轉(zhuǎn)移到1/2-1MS+1g·L活性炭的成苗培養(yǎng)基中可發(fā)育成正常植株,植株室外栽培成活率達(dá)90%。對100株再生植株的染色體數(shù)進(jìn)行分析,發(fā)現(xiàn)3株三倍體(2n=3x=51)、6株四倍體(2n=4x=68)和2株六倍體(2n=6x=102)。關(guān)
