1、Osterix基因敲除及過表達(dá)對小鼠脊椎骨量的影響*陳思旭,宗兆文,賈敏,華祥,沈岳,趙玉峰,郭慶山,劉道城(400042重慶,第三軍醫(yī)大學(xué)大坪醫(yī)院野戰(zhàn)外科研究所創(chuàng)傷科,創(chuàng)傷、燒傷與復(fù)合傷國家重點實驗室)[摘要]目的研究小鼠Osterix基因敲除及過表達(dá)后對其脊椎骨量的影響及作用機(jī)制。方法將12周齡的野生型小鼠(WT)、Osterix基因敲除小鼠(Osterix-KO)和Osterix過表達(dá)轉(zhuǎn)基因小鼠(Osterix-CA)分為3組(每組8只,均為雄性),分別采用X線攝像和HE染色觀察小鼠腰椎骨量的變化,TRAP染色檢測破骨細(xì)胞水平,免疫組織化學(xué)檢測NF-κB受體活化因子配體(RAN
2、KL)、骨涎蛋白(BSP)和骨橋蛋白(OPN)表達(dá)水平,并將所得數(shù)據(jù)進(jìn)行統(tǒng)計學(xué)分析。結(jié)果Osterix-KO小鼠在X線和HE染色表現(xiàn)為腰椎骨密度和骨量增加,破骨細(xì)胞數(shù)量顯著減少(P<0.05),椎體中RANKL、BSP和OPN的表達(dá)水平顯著降低(P<0.05),而Osterix-CA小鼠的脊椎X線、HE染色表現(xiàn),及破骨細(xì)胞數(shù)量與WT小鼠無明顯差異。結(jié)論Osterix基因敲除小鼠脊柱骨吸收活性減弱伴成骨細(xì)胞分化能力降低,進(jìn)而發(fā)生骨量明顯增加。[關(guān)鍵詞]Osterix;脊椎;骨量[中圖法分類號][文獻(xiàn)標(biāo)志碼]AEffectTheroleofOsterixosterixknock-outo
