1、幼鼠癲癇持續(xù)狀態(tài)成年后腦內(nèi)髓鞘變化的研究劉淑丹1,張桂禎2,王晗知1,何揚濤1,肖嵐1,李紅麗1(400038重慶,第三軍醫(yī)大學(xué)基礎(chǔ)醫(yī)學(xué)部組織學(xué)與胚胎學(xué)教研室1,610083成都,成都軍區(qū)總醫(yī)院信息科2)[摘要]目的探討致癲癇持續(xù)狀態(tài)幼鼠(statusepilepticus,SE)成年后發(fā)展為自發(fā)反復(fù)性癲癇發(fā)作(spontaneousrecurrentepilepticseizure,SRS)的大鼠腦內(nèi)髓鞘的變化。方法采用氯化鋰-匹羅卡品腹腔注射誘P20-P28體重(70±5)g雄性SD幼鼠SE模型,應(yīng)用多道生理信號采集處理系統(tǒng)檢測大鼠腦電圖(electroencephalogra
2、m,EEG)異?;顒硬?,采用Nissl染色方法觀察腦內(nèi)神經(jīng)元的變化,證實SRS模型大鼠誘導(dǎo)成功;應(yīng)用盧卡斯快藍(lán)(luxolfastblue,LFB)染色及髓鞘堿性蛋白(myelinbasicprotein,MBP)免疫組織化學(xué)染色等技術(shù)觀察腦內(nèi)髓鞘的變化。結(jié)果SRS模型組腦電圖與正常對照組相比,可見頻發(fā)尖波、棘-慢、尖-慢綜合波;Nissl染色顯示神經(jīng)元數(shù)目在海馬CA1(242.1±33.6)、CA3(354.8±26.8)和齒狀回門區(qū)(61.8±8.3)減少(P<0.01),部分細(xì)胞變性和壞死。LFB染色可見SRS模型組在胼胝體(callosumcorpus,cc;0.162±0
3、.017)、扣帶回(cingulatedgyrus,cg;0.139±0.013)與海馬白質(zhì)(0.080±0.007)區(qū)域內(nèi)的著色范圍明顯縮小,著色淺淡,與正常對照組相比有顯著差異(P<0.01);MBP免疫陽性沉淀在cc(0.183±0.013)、cg(0.192±0.016)、第一軀體感覺皮質(zhì)區(qū)(S1BF;0.076±0.012)和第一聽皮質(zhì)區(qū)(Au1;0.085±0.014)內(nèi)的平均光密度值(OD)顯著低于正常對照組(P<0.01)。結(jié)論致癇幼鼠成年后伴有慢性癲癇發(fā)作可造成中樞神經(jīng)系統(tǒng)髓鞘脫失。[關(guān)鍵詞]氯化鋰-匹羅卡品;慢性癲癇;腦電圖;脫髓鞘;髓鞘堿性蛋白[中圖法分類號]
