1、新型重組高效復(fù)合干擾素對(duì)卵巢癌細(xì)胞生長(zhǎng)抑制及促凋亡作用【摘要】目的:探討新型重組高效復(fù)合干擾素對(duì)卵巢癌HO8019細(xì)胞的作用,并與同類型復(fù)合干擾素、普通重組Ⅰ型干擾素、化療藥物對(duì)比。方法:應(yīng)用新型重組高效復(fù)合干擾素、復(fù)合α干擾素、干擾素α-2b和順鉑以0.156ug/ml、0.625ug/ml、2.5ug/ml及10ug/ml濃度作用于體外培養(yǎng)的HO8019細(xì)胞72小時(shí)后,MTT法檢測(cè)并計(jì)算細(xì)胞抑制率并比較。應(yīng)用新型重組高效復(fù)合干擾素、復(fù)合α干擾素、干擾素α-2b和順鉑以0.156ug/ml、0.625ug/ml、2.5ug/ml濃度作用于體外培養(yǎng)的HO8
2、019細(xì)胞72小時(shí)后,以流式細(xì)胞儀檢測(cè)HO8019細(xì)胞的凋亡率并進(jìn)行比較。結(jié)果:幾種干擾素及順鉑均能導(dǎo)致HO8019細(xì)胞生長(zhǎng)抑制及促進(jìn)細(xì)胞凋亡。相同濃度條件下,新型重組高效復(fù)合干擾素和順鉑對(duì)HO8019細(xì)胞的抑制及凋亡作用強(qiáng)于復(fù)合α干擾素和干擾素α-2b的作用,尤以順鉑的作用最強(qiáng)。結(jié)論:重組高效復(fù)合干擾素對(duì)體外培養(yǎng)的卵巢癌HO8019細(xì)胞的生長(zhǎng)有抑制作用,并能促進(jìn)HO8019細(xì)胞凋亡;作用雖然不及化療藥物順鉑,但較同類型干擾素、普通重組Ⅰ型干擾素的作用強(qiáng)?!娟P(guān)鍵詞】干擾素;卵巢癌;基因重組TheGrowthInhibitionand10Αpoptosis-

7、vicacancercellinvitro.TheeffectisinferiortoDDP,butbetterthanthesametypeinterferonandgeneraltypeIinterferon.【Keywords】Interferon;CervicaIcancer;Generecombination干擾素(interferon,IFN)是較早發(fā)現(xiàn)和應(yīng)用的一類細(xì)胞因子,不僅具有抗病毒復(fù)制、抑制細(xì)胞分裂、誘導(dǎo)細(xì)胞分化、增強(qiáng)細(xì)胞吞噬功能等作用,還可以調(diào)節(jié)免疫反應(yīng),誘導(dǎo)細(xì)胞特定基因的表達(dá),加強(qiáng)機(jī)體在抗腫瘤方面的免疫應(yīng)答[1-3]。其中以a-干擾
