1、Bim1基因在胃癌中的表達及其臨床意義作者：祁衛(wèi)東,陳鎖成,王勝,許文榮,許曉蒙,孫靖【摘要】目的：探討B(tài)im1基因在胃腺癌細胞株、胃癌組織和癌旁組織以及胃癌患者外周血中的表達水平及其與臨床病理特征的關系。方法:收集80例胃癌患者的癌組織和癌旁組織,培養(yǎng)胃腺癌細胞株(SGC7901,BGC823,AGS),采用RTPCR和實時PCR的方法檢測Bmi1基因的表達情況。另外同時收集Bim1基因陽性胃癌患者外周血標本以及健康體檢者外周血標本各20例,用淋巴細胞分離液(Ficoll)分離獲得單個核細胞,采用巢式PCR和實時PCR的方法檢測其Bmi

2、1基因的表達情況。采用蛋白質(zhì)印跡法驗證胃癌組織和對應的癌旁組織以及胃癌患者及健康體檢者血單個核細胞中Bim1蛋白的表達情況。結(jié)果:胃腺癌細胞株(SGC7901,BGC823,AGS)中Bim1均高表達;胃癌組織中Bim1表達(85%)明顯高于癌旁組織(30%)(P<0.05);胃癌患者外周血單個核細胞Bim1基因的陽性率為60%,顯著高于健康體檢者(P<0.05)。胃癌組織和癌旁組織中Bim1的蛋白水平存在差異,而在外周血中未見明顯差異。結(jié)論:Bim1基因在胃腺癌細胞株以及胃癌組織中高表達,胃癌患者外周血單個核細胞Bim1的
