1、幽門螺桿菌外膜36kDa蛋白重組表達與初步鑒定幽門螺桿菌外膜36kDa蛋白重組表達、純化及初步鑒定鞠小麗,邵世和,郝渭濱,董蘇榮,崔蕾蕾摘要:[目的]構(gòu)建幽門螺桿菌(H.pylori)NCTC11637菌株36kDa(OMP36)外膜蛋白基因原核表達系統(tǒng),探索研制H.pylori疫苗的新途徑。[方法]培養(yǎng)和收集H.pyloriNCTC11637菌株,提取基因組DNA,PCR擴增目的基因片斷。將其克隆到T載體并測序,再將目的基因克隆入PET32a(+),構(gòu)建重組表達載體PET32a-OMP36。轉(zhuǎn)化E.coliBL21后IPTG誘導表達,經(jīng)S
2、DS-PAGE鑒定、鎳親和層析純化、Westernblot檢測表達蛋白。[結(jié)果]測序分析表明,插入的基因片斷為987bp,表達的蛋白的相對分子質(zhì)量(Mr)約為36kDa,并顯示具有良好的抗原性。SDS-PAGE檢測表明,表達量占細菌總蛋白的37%,鎳親和層析純化率約92%。Westernblot檢測表達蛋白具有良好的抗原活性。[結(jié)論]成功克隆了外膜蛋白OMP36kDa的編碼基因,其表達產(chǎn)物OMP36有望成為新的Hp疫苗候選分子,為H.pylori疫苗的研制和試劑盒的開發(fā)奠定了良好的基礎(chǔ)。關(guān)鍵詞:幽門螺桿菌;外膜蛋白;基因表達;蛋白純化EXP
