1、雷公藤多甙對(duì)急性移植物抗宿主病小鼠的作用作者:于艷秋,張義俠,金玉楠,盧曉梅,王巍,張海鵬【摘要】本研究應(yīng)用雷公藤多甙防治小鼠急性移植物抗宿主病。對(duì)受致死量照射的C57BL/6受鼠,注入供鼠BABC/c骨髓與脾淋巴細(xì)胞的混合液,并給予雷公藤多甙、環(huán)孢素A、甲氨蝶呤處理。用免疫組織化學(xué)法、流式細(xì)胞術(shù)和ELISA法檢測(cè)各組小鼠T淋巴細(xì)胞、黏附分子、細(xì)胞因子。結(jié)果表明:所有異基因?qū)φ战M小鼠,在30天內(nèi)死于aGVHD,而雷公藤多甙組(19/21)、環(huán)孢素A+甲氨蝶呤組(13/21)和雷公藤多甙+環(huán)孢素A組(17/21)的大部分小鼠存活時(shí)間超過了30天,并沒有明顯的aGVHD癥狀表現(xiàn)。雷公
2、藤多甙、環(huán)孢素A、甲氨蝶呤可明顯降低皮膚、肺組織CD3+、CD4+、CD8+、CD11a+、CD18+細(xì)胞(P<0.05)和脾組織CD3+,CD4+,CD8+,CD4+CD11a+,CD4+CD18+,CD8+CD11a+,CD8+CD18+細(xì)胞(P<0.05)。而小腸組織CD3+、CD4+、CD8+細(xì)胞變化不明顯(P>005)。同時(shí),雷公藤多甙可降低受鼠血清中IL-2、TNFα濃度和脾組織IL-2、TNFα的mRNA的表達(dá)(P<0.05);上調(diào)血清中IL-10的水平(P<0.05),但對(duì)IL-4的作用不顯著(P>0.05)。結(jié)論:雷公藤多甙
