1、當(dāng)歸多糖對(duì)輻射損傷小鼠骨髓基質(zhì)細(xì)胞的影響關(guān)雪晶,吳宏,何曉莉,姜蓉(400016重慶,重慶醫(yī)科大學(xué)基礎(chǔ)醫(yī)學(xué)院干細(xì)胞與組織工程研究室,組織胚胎學(xué)教研室)[摘要]目的研究當(dāng)歸多糖(Angelicapolysaccharides,APS)對(duì)輻射損傷小鼠骨髓基質(zhì)細(xì)胞(bonemarrowstromalcell,BMSC)的保護(hù)作用。方法BALB/c小鼠隨機(jī)數(shù)字表法分為10組,每組6只,共計(jì)240只。正常組不作任何處理,其余9組經(jīng)X射線(xiàn)照射后分別給予生理鹽水(NS)、2mg/kgAPS和8mg/kgAPS,給藥至第7、
2、14、21天計(jì)數(shù)小鼠外周血白細(xì)胞(WBC)、紅細(xì)胞(RBC)、血小板(PLT)及骨髓單個(gè)核細(xì)胞(BMNC);觀(guān)察BMSC生長(zhǎng)達(dá)80%貼壁所需時(shí)間及形成的成纖維細(xì)胞集落(CFU-F)數(shù)量;流式細(xì)胞術(shù)檢測(cè)BMSC細(xì)胞周期、凋亡率及表面黏附分子CD54、CD106的表達(dá)水平。結(jié)果與正常組比較,NS組外周血WBC、RBC、PLT及BMNC計(jì)數(shù)明顯減少(P<0.05);BMSC達(dá)80%貼壁所需時(shí)間明顯延長(zhǎng),CFU-F數(shù)明顯減少(P<0.05);G0/G1期細(xì)胞比例顯著增加,S期細(xì)胞比例顯著降低(P<0.05);BMSC
3、細(xì)胞凋亡率明顯增高(P<0.05);CD54、CD106表達(dá)明顯降低(P<0.05)。2mg/kgAPS、8mg/kgAPS組與NS組相比,外周血各指標(biāo)、BMNC數(shù)及CFU-F數(shù)明顯增加(P<0.05);BMSC達(dá)80%融合時(shí)間顯著縮短(P<0.05);G0/G1期比例明顯降低,S期比例顯著增加(P<0.05);凋亡率顯著降低(P<0.05);CD54、CD106表達(dá)明顯增高(P<0.05)。第21天8mg/kgAPS組各指標(biāo)均恢復(fù)正常。結(jié)論APS可能是通過(guò)提高BMSC貼壁及增殖能力,加速BMSC由G0/G1
