2、度的丙泊酚作用神經(jīng)元不同時間。四甲基偶氮唑藍(lán)(M,lTr)法測細(xì)胞存活率,TUNEL染色法測細(xì)胞凋亡指數(shù),Western—blot免疫印記檢測Cleaved—caspase3蛋白的表達。結(jié)果:丙泊酚作用12h后.細(xì)胞存活率在6個組中無明顯變化,作用24h及48h后,較高濃度組細(xì)胞存活率降低趨勢明顯。且較高濃度作用于神經(jīng)元24h后凋亡指數(shù)及Cleaved-caapase3蛋白的表達均呈濃度依賴性增高(戶如.05)。結(jié)論:高濃度長時間丙泊酚作用于大腦皮層神經(jīng)元可降低其存活力和促進細(xì)胞凋亡,這一結(jié)論對臨床兒童麻醉用藥有指導(dǎo)意義?!娟P(guān)鍵詞l丙泊酚;皮層神經(jīng)
