4、earlyseedlingdevelopmentandstomatalmovement.GCR2wasalsoproposedtobeaseven-transmembrane(7TM)G-protein-coupledreceptor(GPCR).HerewecharacterizeGCR2andoneofitstwohomologs,GCR2-LIKE1(GCL1),inABA-mediatedseedgerminationandearlyseedlingdevelopmentinArabidopsis.Weshowthatloss-of-functionmutationsin

5、GCL1didnotconferABAinsensitivity.Similarly,wedidnotobserveABAinsensitivityinthreeindependentgcr2alleles.Furthermore,wegeneratedgcr2gcl1doublemutantsandfoundthatthedoublemutantsstillhadnearwild-typeresponsestoABA.Consistentwiththis,wefoundthatthetranscriptionofABAmarkergeneswasinducedbyABAtole

6、velsthatwerecomparableinwildtypeandgcr2andgcl1singleanddoublemutants.Ontheotherhand,theloss-of-functionallelesofthesoleArabidopsisheterotrimericGproteinasubunit,GPA1,werehypersensitivetoABAintheABA-inhibitionofseedgerminationandearlyseedlingdevelopment,disfavoringageneticcouplingofGCR2byGPA1.

7、Usingmultiplerobusttransmembranepredictionsystems,GCR2waspredictednottobea7TMprotein,astructuralhallmarkofGPCRs.Takentogether,ourresultsdonotsupportthenotionthatGCR2isanABA-signalingGPCRinseedgerminationandearlyseedlingdevelopment.Keywords:7T
