1、·1042·中華實驗外科雜志2014年5月第31卷第5期ChinJExpSurg,May2014,Vo1.31,No.5·實驗研究·蛋白酶體抑制劑MG132增加順鉑對食管鱗癌細胞的殺傷作用楊洋樊玉霞劉東雷吳愷溫豐標趙松【摘要】目的探討蛋白酶體抑制劑MG132聯(lián)合順鉑對食管鱗癌細胞的殺傷作用及其機制。方法分別以順鉑(100mg/L)、MGI32(5p~mol/L)及順鉑+MG132處理食管鱗癌EC9706細胞24h,細胞計數(shù)試劑盒(CCK一8)法檢測細胞活力,膜聯(lián)蛋白V(AnnexinV)一異硫氰酸熒光素(FITC)細胞凋亡檢測試劑盒定量檢測細胞凋亡率,Westernblo
2、t檢測凋亡相關(guān)蛋白半胱氨酰天冬氨酸特異性蛋白酶(Caspase)一8、Caspase一3和核轉(zhuǎn)錄因子(NF)一KB的表達。結(jié)果MG132可以抑制腫瘤細胞的增殖,增強順鉑對食管鱗癌細胞的殺傷作用,使細胞存活率從(68.19±3.41)%降低至(29.37±4.16)%(P<0.05)。MG132聯(lián)合順鉑處理組的細胞凋亡率為(68.45±2.58)%,明顯高于單獨處理組的(23.55±1.23)%和(25.87±2.07)%(P<0.01)。聯(lián)合用藥組細胞Caspase一8、Caspase一3的表達水平高于順鉑處理組,但NF—KB較單獨順鉑處理組明顯下降(P<0.05)。結(jié)論
