2、·L脂肪多糖(IipopoIysaccharide,LPS)的混合組的培養(yǎng)inducedbyhumanHMGBl基分別培養(yǎng)THPI細(xì)胞0,2,4,6,8,I0,I2和24h,同上留BIELiang-feng,CHENGXiao-dong,SUMing-ouan,YUE取離心后的培養(yǎng)液上清,用雙抗體夾心ELISA法測(cè)其上清中oiao-Hong,YUWen-Bin,HAOXiao-ke的TNF-!含量.結(jié)果:將HMGBI加入到單核細(xì)胞培養(yǎng)液中DepartmentofCIinicaILaboratories,XijingHos
3、pitaI,F(xiàn)ourthMiIi-能明顯刺激其TNF-!的分泌.在0.I~50mg·L-I范圍內(nèi),taryMedicaIUniversity,Xi’an7I0033,ChinaHMGBI誘導(dǎo)THPI的TNF-!分泌增加,呈顯著劑量依賴性關(guān)-I系,50mg·LHMGBI能明顯誘導(dǎo)THPI細(xì)胞的凋亡;【Abstract】AIM:ToexplorethesecretionpatternofTNF-!HMGBI組和HMGBI與LPS的混合組THPI的TNF-!分泌在inhumanTHPIcellsinducedbyhumanHM

6、asobservedbyflowcytometry.RESULTS:HMGBItein,HMGBI)可能作為晚期炎癥因子參與了內(nèi)毒素markedlyinducedthereleaseofTNF-!fromTHPIcellsina的致病過(guò)程(Science,I999;248:25I).HMGBI與-I-Idose-dependentmannerfrom0.Img·Lto50mg·LTNF-!,IL-I"等早期細(xì)胞因子相比,出現(xiàn)較晚且持HMGBI.ThereweretwopeaksofTNF-!afterTHPIcells續(xù)
7、時(shí)間更長(zhǎng),可能作為潛在的晚期介質(zhì)參與了膿毒癥wereinducedbyHMGBIaloneormixedwithLPSinthepe-[I]-I的病理生理過(guò)程.鼠重組HMGBI能使人外周血riodof0~24h.50mg·LHMGBImarkedlyeffectedap-[2]單核細(xì)胞分泌促炎細(xì)胞因子TNF-!,但具體機(jī)制和optosisofTHPIcells.CONCLUSION:HumanHMGBIcan[3]markedlyinducethereleaseofTNF-!fromTHPIcellsand規(guī)律尚不明了.
8、故我們采用重組人HMGBI來(lái)刺激effectapoptosisofTHPIcells,indicatingthathuman人單核/巨噬細(xì)胞系THPI,觀察其TNF-!的分泌規(guī)HMGBImayplayanimportantroleintheoccurrenceand律,并且對(duì)其凋亡情況進(jìn)行了研究.developmen
