1、昆明醫(yī)科大學(xué)學(xué)報(bào)2015,36(5):5~7CN53—1221/RJournalofKunmingMedicalUniversity舌鱗癌Cal27裸鼠移植瘤模型的建立及其意義吳京蔓,吳勇(昆明醫(yī)科大學(xué)附屬12腔醫(yī)院口腔頜面外科,云南昆明650031)[摘要]目的為抑癌藥物的臨床研究建立人舌鱗癌動(dòng)物模型.通過模擬與人體內(nèi)環(huán)境及生物學(xué)特性相似的動(dòng)物模型,觀察腫瘤細(xì)胞的生物學(xué)、病理學(xué)行為及轉(zhuǎn)移情況.方法選4~6周BALB/C—nu裸鼠,將體外培養(yǎng)的人舌鱗癌Ca127細(xì)胞接種于裸鼠右腋皮下,觀察裸鼠體質(zhì)量、皮下成瘤情況、腫瘤體積;待裸鼠體質(zhì)量下降至原體質(zhì)量的25%時(shí),用頸椎脫臼法處死裸鼠
2、,剝離瘤體組織進(jìn)行解刨學(xué)觀察及病理學(xué)觀察.結(jié)果實(shí)驗(yàn)裸鼠在接種Ca127細(xì)胞第7~12d后右腋下出現(xiàn)腫瘤,接種濃度為每只0.1mL約2X106個(gè)細(xì)胞,成瘤率為86.7%.HE染色示腫瘤組織呈典型鱗癌表現(xiàn);淋巴結(jié)及主要臟器未見轉(zhuǎn)移.結(jié)論成功建立舌鱗癌Ca127裸鼠移植瘤模型.此動(dòng)物模型能較客觀地反映人舌鱗癌的生物學(xué)行為,為進(jìn)行抑癌藥物的臨床研究與治療打下良好基礎(chǔ).[關(guān)鍵詞]舌鱗癌;Ca127細(xì)胞;裸鼠;移植瘤;動(dòng)物模型[中圖分類號(hào)]R739.8[文獻(xiàn)標(biāo)識(shí)碼]A[文章編號(hào)]2095-610X(2015)05—00O5—04CarcinomaCal27CellsMOdelinNUdeMic
