2、K一8(cellcountingkit一8)和平板克隆形成實(shí)驗(yàn)檢測(cè)A549和A549／CDDP細(xì)胞的自我更新能力，F(xiàn)CM法檢測(cè)A549和A549／CDDP貼壁細(xì)胞及細(xì)胞球中CD90細(xì)胞的比例，細(xì)胞毒性實(shí)驗(yàn)檢測(cè)CD90A549和CD90一A549細(xì)胞對(duì)CDDP的耐藥性。建立CD90A549和CD90一A549細(xì)胞裸鼠移植瘤模型，檢測(cè)體內(nèi)CD90A549細(xì)胞對(duì)CDDP的耐藥性。結(jié)果：與A549細(xì)胞比較，A549／CDDP細(xì)胞的成球能力、細(xì)胞增殖及克隆形成能力更強(qiáng)(P<0．01)。A549／CDDP貼壁細(xì)胞和細(xì)胞球中CD90細(xì)胞比例分別高于A549貼壁細(xì)胞和細(xì)胞球(P<0．05)。
3、CDDP作用CD90A549細(xì)胞和CD90A549細(xì)胞的IC值分別為8．50和1．50gmol／L。經(jīng)CDDP干預(yù)后，CD90A549細(xì)胞裸鼠移植瘤體積大于CD90一A549細(xì)胞(P<0．05)。結(jié)論：A549／CDDP細(xì)胞具有較強(qiáng)的自我更新能力和較高比例的CD90亞群細(xì)胞，CD90’。A549細(xì)胞表現(xiàn)出較高的CDDP耐藥性，提示CD9O可能是肺癌的干細(xì)胞標(biāo)志物?！娟P(guān)鍵詞】肺腫瘤；腫瘤干細(xì)胞；順鉑；抗藥性，腫瘤；CD90；A549細(xì)胞[中圖分類號(hào)】R734．2【文獻(xiàn)標(biāo)志碼】A【文章編號(hào)】1000—7431(2013)09—0770—06Therelationshipbetwe
