2、OV3凋亡及生長(zhǎng)抑制作用。方法噻唑藍(lán)法檢測(cè)Smac小分子compound3單獨(dú)應(yīng)用組、順鉑單獨(dú)應(yīng)用組及二者聯(lián)合應(yīng)用組對(duì)卵巢癌細(xì)胞SKOV3的生長(zhǎng)抑制率、存活率的影響;并繪制生長(zhǎng)曲線。AnnexinV/PI雙標(biāo)流式細(xì)胞術(shù)檢測(cè)Smac小分子compound3單獨(dú)應(yīng)用組、順鉑單獨(dú)應(yīng)用組及二者聯(lián)合應(yīng)用組對(duì)SKOV3凋亡率的影響。Western印跡檢測(cè)Smac小分子compound3單獨(dú)應(yīng)用組、順鉑單獨(dú)應(yīng)用組及二者聯(lián)合應(yīng)用組對(duì)SKOV3的x連鎖凋亡抑制蛋白(X—linkedinhibitorofapoptosisprotein,XIAP)、半胱氨酸一天冬氨酸蛋白
3、酶一3(caspase一3)的表達(dá)影響。結(jié)果Smac小分子compound3單獨(dú)應(yīng)用組、順鉑單獨(dú)應(yīng)用組及二者聯(lián)合應(yīng)用組12、24、48hSKOV3生長(zhǎng)受到抑制,且呈時(shí)間依賴性。二者聯(lián)合應(yīng)用組與順鉑單獨(dú)應(yīng)用組比較生長(zhǎng)抑制率明顯升高。二者聯(lián)合應(yīng)用組的凋亡率高于順鉑單獨(dú)應(yīng)用組及Smac小分子compound3單獨(dú)應(yīng)用組。Smac小分子compound3單獨(dú)應(yīng)用組及二者聯(lián)合應(yīng)用組SKOV3的XIAP表達(dá)降低,二者聯(lián)合應(yīng)用組的caspase一3的激活片段表達(dá)明顯高于其他兩組。結(jié)論Smac小分子compound3能夠調(diào)節(jié)順鉑對(duì)SKOV3生長(zhǎng)抑制作用,促進(jìn)順鉑對(duì)卵巢
