2、ED1單克隆抗體免疫熒光細(xì)胞化學(xué)鑒定KCs;顯微鏡下觀察KCs形態(tài)變化。結(jié)果:獲取的KCs數(shù)量為(2.41±0.32)×10/只,其中活細(xì)胞數(shù)量占(92.3±2.12)%;吞噬實(shí)驗(yàn)顯示(95.2±2.58)%的細(xì)胞內(nèi)存在碳素顆粒;免疫熒光化學(xué)檢測(cè)證明KCs純度為(96.3-4-1.46)%;在顯微鏡下觀察KCs形態(tài),36h細(xì)胞形態(tài)變得不規(guī)則,3d后呈星形或多角形,體外培養(yǎng)可以存活7—10d。結(jié)論:此種KCs分離方法操作相對(duì)簡(jiǎn)便,獲取的細(xì)胞數(shù)量、活性功能、純度等方面均能達(dá)到進(jìn)一步的實(shí)驗(yàn)要求,值得推廣。關(guān)鍵詞Kupffer細(xì)胞;細(xì)胞分離;細(xì)胞

8、(KCs)是機(jī)體單核一巨噬細(xì)胞系統(tǒng)的最兩步原位灌注法結(jié)合Precoll密度離心法成功分離了大鼠KCs,大組成分,數(shù)量約占80%一90%,在機(jī)體免疫、內(nèi)環(huán)境穩(wěn)定其純度活性功能均達(dá)到實(shí)驗(yàn)要求,為相
