1、產(chǎn)漆酶裂褶菌篩選和其產(chǎn)酶培養(yǎng)條件優(yōu)化探究 摘要:從采自秦嶺的森林腐木菌苔上分離到一株產(chǎn)漆酶的裂褶菌(SchizophyllumcommuneFr.)菌株mys005,對其產(chǎn)漆酶培養(yǎng)方式、培養(yǎng)基組成及培養(yǎng)條件進(jìn)行了優(yōu)化試驗(yàn)。結(jié)果表明,裂褶菌菌株mys005的產(chǎn)漆酶適宜培養(yǎng)方式為液體淺層振蕩培養(yǎng),含有Cu2+、Mn2+、Zn2+的微量元素復(fù)合液、洋蔥(AlliumcepaL.)、麥(TriticumaestivumL.)麩對其產(chǎn)漆酶都有較大的促進(jìn)作用,而愈創(chuàng)木酚則對其生長和產(chǎn)漆酶有抑制作用。由新鮮馬鈴薯(SolanumtuberosumL.)200g、K2HPO4
2、1.00g、NaCl0.50g、MgSO4·7H2O0.50g、NaNO32.50g、CaCl2·2H2O0.10g、FeCl30.02g、吐溫-801.20mL溶于1000mL去離子水中組成的培養(yǎng)基在添加微量元素復(fù)合液(50mg/mLCuCl2·2H2O+20mg/mLMnSO4·H2O+10mg/mLZnSO4·7H2O)3mL、洋蔥100g、麥麩10g后,并且培養(yǎng)基起始pH4.5、培養(yǎng)溫度30℃、培養(yǎng)搖床轉(zhuǎn)速200r/min、培養(yǎng)時(shí)間7d,則搖瓶發(fā)酵的漆酶產(chǎn)酶量最高可達(dá)1121U/mL,顯示出了良好的工業(yè)開發(fā)潛力。關(guān)鍵詞:漆酶;裂褶菌(Schizophyl
