3、產(chǎn)纖維素酶菌株進(jìn)行初篩,選取透明圈較大的菌株進(jìn)行搖瓶發(fā)酵復(fù)篩,根據(jù)菌株形態(tài)、生理生化特征對(duì)菌株進(jìn)行初步鑒定,通過正交法優(yōu)化產(chǎn)酶條件。結(jié)果表明,經(jīng)初篩和復(fù)篩得到1株酶活相對(duì)較高的菌株B-12,經(jīng)初步鑒定為芽孢桿菌屬(Bacillussp.)。1.25%麥芽浸粉為碳源、1.5%KNO3為氮源、0.2%的NaCl、0.1%的CMC-Na、接種量6%、培養(yǎng)時(shí)間44h為B-12產(chǎn)酶的適宜條件。優(yōu)化后發(fā)酵液中的內(nèi)切葡聚糖酶活(CMCA)為111.710U/mL,較培養(yǎng)44h后的酶活提高了8.78%;濾紙酶活(FPA)為35.017U/mL,提高了387.
