1、摘要糙皮側(cè)耳P拓“陽n姆D5艦口f酣(Jacq.ex.Fr)QueI(簡稱PO)系擔(dān)子菌綱口蘑科真菌,近年來學(xué)者們側(cè)重于對(duì)其化學(xué)成分、藥理作用、優(yōu)化培育等方面的研究。弄清其結(jié)構(gòu),對(duì)于深入了解糙皮側(cè)耳的藥理活性,更好地利用糙皮側(cè)耳這~廣泛又具有多種功能的真菌資源是至關(guān)重要的。本文提純了PO水溶性多糖并研究了其結(jié)構(gòu),為糙皮側(cè)耳的藥理作用和臨床應(yīng)用提供理論依據(jù)。PO經(jīng)熱水煮提、醇析,分離出水溶性粗多糖。Gc分析表明粗多糖的單糖組成為葡萄糖(Glc)、甘露糖(Man)、半乳糖(Gal)、巖藻糖(Fuc),摩爾比依次為:3.88:2.28:3.28:1.
2、oo,說明PO粗多糖為一種中性雜多糖。PO粗多糖經(jīng)凍融離心、鏈蛋白酶酶解、sevag法脫蛋白、乙醇分級(jí)、凝膠柱層析方法純化得水溶性多糖POP。,凝膠柱層析、比旋光度測定、高效液相色譜(HPLc)分析證明POR組分平均分子量為2.4萬,酚一硫酸法測定總糖含量為95%,氣相色譜(GC)分析其單糖組成為:Glc、MaIl、Gal、Fuc,摩爾比為:17.50:4.42:39.88:1.oo。HPLc、比旋光度測定等方法,檢驗(yàn)其均一陛。結(jié)果表明POP。分子量及極性均為均一組分。POP3經(jīng)部分酸水解、高碘酸氧化、smith降解、甲基化、Gc—Ms分析、I
