2、研究健脾理氣化濕方對(duì)大鼠脂肪肝的保護(hù)作用。方法:雄性Wistar大鼠40只,隨機(jī)分正常組、模型組、低劑量組、高劑量組。模型組、低劑量和高劑量組以40%CCl4橄欖油溶液1ml/kg體質(zhì)量腹腔注射,2次/周,共4周。模型組、低劑量和高劑量組在第1、2周給予高脂低蛋白飼料,第3、4周予純玉米粉飼料。自造模之日起,按照1ml/100g大鼠體重給予低劑量和高劑量中藥灌胃。實(shí)驗(yàn)結(jié)束后大鼠經(jīng)下腔靜脈采血并分離血清,統(tǒng)一于肝臟左葉切取組織,于10%中性福爾馬林固定。檢測(cè)血清ALT、AST、GGT活性,肝組織行HE染色。結(jié)果:模型組大鼠血清AL
3、T、AST和GGT均較正常組顯著增高(P<0.01);經(jīng)健脾理氣化濕方中藥干預(yù),ALT、AST和GGT均顯著下降(P<0.05),其中高劑量組ALT和AST下降幅度均優(yōu)于低劑量組(P<0.05),而低劑量組和高劑量組GGT下降幅度無(wú)統(tǒng)計(jì)學(xué)意義(P>0.05)。HE染色可見(jiàn)模型組大鼠小葉結(jié)構(gòu)紊亂,肝細(xì)胞胞質(zhì)疏松,肝細(xì)胞變性,胞漿內(nèi)見(jiàn)大量脂肪空泡。健脾理氣化濕方干預(yù)組可見(jiàn)大鼠肝細(xì)胞損傷及脂肪變性程度明顯減輕,肝小葉結(jié)構(gòu)比較清晰,肝細(xì)胞胞漿內(nèi)脂肪空泡與模型組相比有顯著差異,尤其以高劑量組改善程度更為明顯。結(jié)論:健脾理氣化濕方對(duì)大鼠脂肪
