1、布魯氏菌S2WboA基因缺失株的構(gòu)建及免疫效果摘要:【目的】WboA基因編碼光滑型布魯氏菌脂多糖(LPS)O-側(cè)鏈合成必須的糖基轉(zhuǎn)移酶,該基因的缺失或破壞會(huì)影響布魯氏菌光滑型表型的形成。本研究擬構(gòu)建WboA基因缺失的重組粗糙型布魯氏菌,以使布魯氏菌弱毒疫苗與野毒株布魯氏菌感染相區(qū)分?!痉椒ā恳载i源光滑型布魯氏菌S2株為研究對(duì)象,通過同源重組將氯霉素抗性基因(Cmr)完全替代S2株的WboA基因,獲得重組粗糙型布魯氏菌rS2-WboA株。【結(jié)果】rS2-WboA株在適宜的培養(yǎng)基(TSA)上傳50代后仍保持對(duì)氯霉素的抗性。用1×107CFUrS2-WboA免疫Balb/c小鼠和豚鼠,1個(gè)
