6、enes,Tat【摘要】目的:構(gòu)建人抗HBsAg單鏈抗體Tat蛋白轉(zhuǎn)導(dǎo)結(jié)構(gòu)域(ScFvTat)融合基因,在大腸桿菌中進(jìn)行表達(dá),并分析ScFvTat融合蛋白的活性及內(nèi)化情況.方法:設(shè)計(jì)引物擴(kuò)增人抗HBsAg單鏈抗體基因,克隆入含有蛋白轉(zhuǎn)導(dǎo)結(jié)構(gòu)域Tat序列的原核表達(dá)載體pTATHA,在大腸桿菌BL21(DE3)LysS內(nèi)誘導(dǎo)融合蛋白表達(dá).表達(dá)產(chǎn)物用SDSPAGE及Westernblot鑒定,用親和層析柱純化.間接ELISA和ELISA競(jìng)爭(zhēng)抑制實(shí)驗(yàn)分析ScFvTat融合蛋白的親和活性,將純化蛋白與HBsAg陽(yáng)性或陰性肝癌細(xì)胞共孵育后,免疫細(xì)胞化學(xué)染色分析ScFvTat融合蛋白的結(jié)合及
7、內(nèi)化情況.結(jié)果:成功地構(gòu)建了人抗HBsAg單鏈抗體Tat融合蛋白的原核表達(dá)載體,在IPTG誘導(dǎo)下獲得了表達(dá),表達(dá)的ScFvTat融合蛋白具有與HBsAg結(jié)合的活性,并且能夠特異性內(nèi)化入HBsAg陽(yáng)性肝癌細(xì)胞.結(jié)論:?jiǎn)捂溈贵w與Tat蛋白轉(zhuǎn)導(dǎo)結(jié)構(gòu)域融合后,實(shí)現(xiàn)了ScFvTat融合蛋白的特異性內(nèi)化,為該單鏈抗體的進(jìn)一步應(yīng)用奠定了基礎(chǔ).【關(guān)鍵詞】單鏈抗體;肝炎表面抗原,乙型;內(nèi)化;基因,Tat0引言單鏈抗體(SeFv)及其與效應(yīng)分子的融合體能否高效地內(nèi)化入細(xì)胞對(duì)于其應(yīng)用非常關(guān)鍵.Tat是新近發(fā)現(xiàn)的一
