2、照,并觀察計(jì)算細(xì)胞純度。以低糖缺氧環(huán)境模擬體內(nèi)缺氧缺血狀態(tài),建立大鼠02A體外缺氧模型,觀察02A在各個(gè)時(shí)間點(diǎn)的形態(tài)學(xué)變化,并開展Westernblot和免疫組化實(shí)驗(yàn),利用激光共聚焦顯微鏡測定熒光強(qiáng)度,反映Tau蛋白量的變化。結(jié)果:純化后02A細(xì)胞胞體呈圓形,有單極或雙極突起,形成克隆球,A285標(biāo)記陽性,免疫熒光鑒定細(xì)胞純度可達(dá)85%以上。隨著暴露在低糖缺氧環(huán)境中時(shí)間的延長,02A細(xì)胞數(shù)量逐漸減少,突起逐漸縮短,免疫熒光強(qiáng)度和灰度值均逐漸減弱,Tau蛋白量呈減少趨勢。結(jié)論:1.通過體外進(jìn)行純化、培養(yǎng),可以獲得較高純度的02A細(xì)胞;2.隨著低糖缺氧時(shí)間的延長,02A
