2、用該體系成功檢測(cè)了150個(gè)轉(zhuǎn)基因柑橘單株系外源基因整合拷貝數(shù)。結(jié)果表明,轉(zhuǎn)基因柑橘中外源基因整合最低拷貝數(shù)為1,最高達(dá)5個(gè),其中1、2和≥3拷貝的單株數(shù)分別占總數(shù)的40.0%、18.0%和42.0%;采用Southernblot技術(shù)進(jìn)行檢測(cè),結(jié)果基本一致,極少數(shù)單株經(jīng)實(shí)時(shí)熒光定量PCR分析的拷貝數(shù)比Southernblot檢測(cè)的數(shù)值高。說明本研究建立的實(shí)時(shí)熒光定量PCR方法檢測(cè)轉(zhuǎn)基因柑橘外源基因整合拷貝數(shù)更加準(zhǔn)確可靠??截悢?shù)為1~2的轉(zhuǎn)基因株系可作為將來開展轉(zhuǎn)基因目標(biāo)性狀研究及轉(zhuǎn)基因生物安全性評(píng)價(jià)有利用價(jià)值的試驗(yàn)材料。關(guān)鍵詞:柑橘;實(shí)時(shí)熒光定量PCR;轉(zhuǎn)基因;拷貝數(shù);So
