2、方法分離桃果實(shí)PpPFK基因的cDNA全長(zhǎng)序列,采用RT-qPCR(reversetranscriptionreal-timequantitativePCR)技術(shù)分析桃果實(shí)在0℃、5℃冷藏過程中目的基因轉(zhuǎn)錄水平的變化。試驗(yàn)成功獲得了全長(zhǎng)1405bp的PpPFK基因序列,該序列具有完整的開放閱讀框(ORF,openreadingframe),編碼232個(gè)氨基酸。對(duì)ORF進(jìn)行PCR的驗(yàn)證結(jié)果和電子克隆的結(jié)果一致。序列分析顯示該基因的氨基酸序列同大豆和葡萄具有很高的相似度。RT-qPCR分析顯示,5℃貯藏下的桃果實(shí)PpPFK基因轉(zhuǎn)錄水平在前14d高于0℃果實(shí),這可能導(dǎo)致5℃貯藏
