1、北京林業(yè)大學本科畢業(yè)論文抗植物毒素Pro.R單域抗體的五聚體構建、表達及生物活性鑒定生物技術03-2李阿茜指導老師閻錫蘊張德強摘要目的:建立單域抗體多聚化的技術平臺,提高其與抗原的親和力,以期實現單域抗體對毒素的高靈敏檢測。方法和結果:從質粒pCANTAB5ES1中PCR擴增出特異識別植物毒素Pro.R的單域抗體S1的基因,亞克隆到五聚體載體pVT2中,構建重組表達質粒pVT2S1,再轉化到E.coliTG1中。經westernblot鑒定了其正確表達,IPTG大量誘導后通過Ni-his親和層析純化,300mM咪唑溶液冼脫蛋白,獲得了高表達的可溶性目的蛋白——pVT2S1。SDS—PAGE
2、分析顯示表達的蛋白區(qū)帶分子量約22kDa左右,與預期相符。ELISA結果表明,純化的pVT2S1蛋白可特異性識別抗原Pro.R。由于pVT2載體上為5×his標簽,與常用表達載體上6×his標簽檢測系統(tǒng)不同,為了統(tǒng)一單體VHH和五聚體VHH的檢測標準,又進行了載體的改造,分別構建了pVT2S1(6×his)、空載體pVT2(6×his)和pETS1(單域抗體,N端為6×his標簽),這些都為進一步研究VHH抗體的生物活性以及實際應用,如抗原的檢測等奠定了較好的基礎。結論:成功建立單域抗體多聚化的技術平臺,實現了載體的統(tǒng)一檢測標準,為高靈敏度檢測毒素Pro.R奠定基礎。關鍵詞:Pro.R,單

7、uchastestingofantigen.Conclusion:WehavesuccessfullysetuptheplatformforscreeningVHHantibody,standardizedthedetectiontagofpentamerandmonomer,andestablishedthefoundationoftoxinhighsensitivitydetection.Keywords:Pro.R
