2、lyzed.E2hascomplexandmannose-rich-typeoligosaccharideside-chains,whichareattachedbyN-glycosidiclinkagestothepolypeptide.GlycosylationofE2isinitiatedattheco-translationallevel,anditisinhibitedbytunicamycin,2-deoxy-glucose,and2-deoxy-2-fluoro-glucose.Thus,E2belongstoaglycoproteintypefoundin
3、manyotherenvelopedviruses.El,incontrast,representsadifferentclassofglycoprotein.Thefollowingobservationsindicatethatitscarbohydrateside-chainshaveO-glycosidirlinkage.(1)TheconstituentsugarsofElare,V-acrtylglucosamine,N-acet,ylgalactosamine,galactose,andneuraminicacid:mannoseandfucoseareab
4、sent.(2)Theside-chainscanberemovedby,!-elimination.(3)GlycosylationofElisnotsensitivetothecompoundsinterferingwith,Vglycosylation.ElisthefirstviralglycoproteinanalyzedthatcontainsonlyO-glycosidiclinkages.Coronavirusesarethereforeasuitablemodelsystemtostudybiosynthesisandprocessingofthisty
5、peofglycoprotein.1.IntroductionCoronavirusesareagroupofpositivestranded,RNA-containingviruses,whichcauserespiratory.gastroentericandcentralnervousdisordersinhumansandanimals(McIntosh.1974;TyrrelletaE.,1978;Robb&Bond,1979;Nagashimaet~1..1978,1979).Gerdesetal.(1981)havereportedserologicalcr
6、oss-reactivitybetweenvirusesisolatedfrommultiplesclerosispatientsandmousehepatitisvirusA59.aprototypecoronavirusthathasbeencharacterizedinsomedetailatthemolecularlevel.Itcontainsthreestructuralproteins:anucleocapsidproteinN(JZ,50.000).whichisphosphorylated(Stohlman&Lai.1979),andthetwoenve
7、lopeglycoproteinsElandE2.GlycoproteinElisprobablyatransmembranalmatrixproteinwiththecarbohydratemoietiesresidingontheoutsideofthevirusparticle(Sturman,1977).E2appearsinthematurevirionintwoforms(M,180,000and~~1,90,000),bothofwhichconstitutetheclub-shapedpeplomersthat
