2、影響,并探討其作用機(jī)制。方法:體外培養(yǎng)人乳腺癌BT549細(xì)胞株,分別加入不同濃度的PN、DDP及PN+DDP至細(xì)胞中,作用48h后,分別采用CCK8細(xì)胞增殖實(shí)驗(yàn)檢測細(xì)胞存活率,流式細(xì)胞儀檢測細(xì)胞周期。結(jié)果:隨著PN工作液濃度的升高,BT549細(xì)胞48h的細(xì)胞存活率逐漸降低。同樣,隨著DDP工作濃度的升高,BT549細(xì)胞48h的細(xì)胞存活率也逐漸下降。并且相應(yīng)濃度的PN和DDP聯(lián)合作用于BT549細(xì)胞48h的存活率降低更加明顯,且差異具有統(tǒng)計(jì)學(xué)意義(P<0.05)。流式細(xì)胞儀測細(xì)胞周期實(shí)驗(yàn)發(fā)現(xiàn),這兩種藥物主要通過阻滯細(xì)胞周期G1,S期的轉(zhuǎn)化。
