1、抗sHLA-I免疫磁珠的制備與鑒定[摘要]目的制備抗sHLA-I免疫磁珠去除紅細(xì)胞懸液和機(jī)采血小板中的sHLA-I分了。方法將單克隆抗體W6/32以共價(jià)偶聯(lián)的方式包被制備免疫磁珠,以FITC標(biāo)記的羊抗鼠二抗標(biāo)記磁珠,用流式細(xì)胞儀鑒定包被效果。取10例紅細(xì)胞懸液和機(jī)采血小板上清和制備好的免疫磁珠孵育后移去磁珠,用ELISA檢測(cè)吸附前后sHLA-I濃度變化評(píng)價(jià)吸附效果。結(jié)果制備的抗sHLA-I免疫磁珠包被效率約為75%o對(duì)紅細(xì)胞懸液和機(jī)采血小板中sHLA-T的吸附效果可達(dá)到84%和83%。結(jié)論采用單克隆抗體W6/32制備的免疫磁珠能有效去除紅細(xì)胞懸液和機(jī)采血小板中的sHLA-I分

7、blehumanleukocyteantigensT(HLAT);Immunemagneticbead儲(chǔ)存血上清中含有高濃度的可溶性組織相容性抗原I類分了sHLA-I,研究表明可溶性組織相容性抗原I類分子sHLA-I與受血者免疫功能抑制(Transfusion-relatedimmunomoduLation,TRIM)的發(fā)生密切相關(guān)[1],常規(guī)的白細(xì)胞過濾技術(shù)無法有效地去除儲(chǔ)存血中的sHLA-I,筆者嘗試采用制備抗sHLA-I免疫磁珠以特異性地吸附去除儲(chǔ)存血中的sHLA-I分子,現(xiàn)報(bào)道如下。1材料與方法
