1、基于RNA-Seq絨山羊皮膚兩種毛囊差異表達(dá)基因的分析蘇蕊喬賢范一星李曉凱張磊馬宇浩王瑞軍王志英劉志紅張燕軍李金泉內(nèi)蒙古農(nóng)業(yè)大學(xué)動(dòng)物科學(xué)學(xué)院動(dòng)物遺傳育種與繁殖自治區(qū)重點(diǎn)實(shí)驗(yàn)室為了研究絨III羊皮膚初級(jí)毛囊和次級(jí)毛囊之間的表達(dá)基因模式的差異,篩選影響毛囊發(fā)育的重要基因,對(duì)6只內(nèi)蒙古絨山羊阿爾巴斯型個(gè)體進(jìn)行RNA-seq測(cè)序,利用生物信息學(xué)方法對(duì)轉(zhuǎn)錄組數(shù)據(jù)進(jìn)行分析,在測(cè)序結(jié)果質(zhì)量控制后的基礎(chǔ)上,對(duì)差異表達(dá)基因進(jìn)行篩選、功能注釋和富集分析。共篩選到617個(gè)在初級(jí)毛囊和次級(jí)毛囊中差異表達(dá)的基因,其中,上
2、調(diào)表達(dá)基因297個(gè),下調(diào)表達(dá)基因320個(gè)。G0功能注釋結(jié)果顯示,上調(diào)表達(dá)基因主要行使電子傳遞鏈、呼吸鏈電子傳遞、線粒體電子傳遞、基因表達(dá)等生物學(xué)功能;K調(diào)表達(dá)基因主要行使細(xì)胞吞噬、蛋0質(zhì)復(fù)性、中性粒細(xì)胞聚集、細(xì)菌真菌防御反應(yīng)等生物學(xué)功能。利用KEGG數(shù)據(jù)庫(kù)作為參考,這些基因主耍參與半胱氨酸和蛋氨酸代謝、RNA聚合、RNA可變剪切、底物磷酸化等通路。PPP2R2A和CDK2AP1基因可能在皮膚毛囊發(fā)育中發(fā)揮重要作用,以上結(jié)果為今后深入研究皮膚毛囊發(fā)育相關(guān)基因及功能提供了依據(jù)。關(guān)鍵詞:絨山羊;毛囊;

7、edgenesare:electrontransportchain,respiratorychainelectrontransport,mitochondrialelectrontransport,geneexpressionandotherbiologicalfunctions.Howeverdown-regulatedexpressionofgenesismainlyexercisedasfollowing:cellautophagy,proteincomplex,neutrophilagg
8、regation,bacterialfungaldefensereactionandotherbiologicalfunctions.UsingKEGGdatabaseasareference,thesegenesaremainlyinvolvedinthemetabolismofcysteineandmethionine,RNAaggregation,shear,Parkinson’sdisease,substratephosphorylation,andAlzheimer’sdisease.
