2、endocardium,andthecoronaryendotheliumduringharvestingandstoragecancompromiseoutcomesafterhearttransplantation.Safeguardingofstructureandfunctionofcardiomyocytesandendotheliumindonorheartsmayleadtoimprovedpatientsurvivalaftertransplantation.Informationgainedfromporcineheartsstoredinstandardt

3、ransplantsolutionwasusedtodesignasuperiorpreservationsolutionthatwouldoptimallyprotectandmaintainorgansfrombeatingheartand/ornonbeatingheartdonorsduringlong-termstorage.MethodsandResults—Multiphotonmicroscopywasusedtoimagedeepwithincardiacbiopsiesandcoronaryarterytissueharvestedfromporcineh

4、eartsobtainedfrombeatingheartandnonbeatingheartdonorsforanalysisofmyocyteandendothelialcellstructureandfunction.Cellstructuralintegrityandviability,calciummobilization,andnitricoxidegenerationweredeterminedwithfluorescenceviabilitymarkers,immunofluorescence,andWesternblots.Duringhypothermic

5、storageinstandardpreservationsolution,Celsior,myocyte,andendothelialviabilitywasmarkedlyattenuatedinheartsobtainedfrombeatingheartdonors.Incontrast,heartsfrombeatingandnonbeatingheartdonorsstoredinthenewlyformulatedSomahsolutiondemonstratedanincreaseinhigh-energyphosphatelevels,protectionof

6、cardiacmyocyteviability,mitochondrialmembranepolarization,andstructuralproteins.Similarly,coronaryarteryendothelialorganizationandfunction,calciummobilization,andnitricoxidegenerationwerewellmaintainedduringtemporalstorageinSomah.Conclusions—TheCelsiorpreservationsolutioninclinicalusetodayh
