4、【Objective】Thepresentstudywasconductedtostudytheexpressionofdifferentialgenesinsugarcaneresponsetowaterstressanditsmolecularmechanisminordertoprovidescientificbasisfordevelopinggeneticimprovementofsugarcanedroughtresistantintegrity.【Method】SugarcanecultivarGTl
5、1wasusedasmaterialtodesignthreetreatmentsviz.contr01.droughtandrewatering.The+1“l(fā)eavesofsugarcaneforthreetreatmentswereusedforextractingtotalRNAtoconstructtheirequalquantitymixedsolutions,andthegeneticexpressingprofilesofcuhivarGTl1atelongationstageunderwaters
6、tresswereconstructedbyusingeDNA-SCoTdifferentialdisplay.Thetranscriptderivedfragments(11DFs)werescreenedandisolatedtosequencing,followedbytheBLASThomologysearchinginNCBIdatabasetodeducegenefunctionbasedonhomologicalgenes.【Result】The180TDFswerescreenedbyusing42
7、primersofSCoTmarker,ofwhich100TDFswereofhighersimilaritywiththeaccessedgenesinNCBIdatabase.Accordingtohomologousgenefunction,these100TDFscouldbeclassifiedinto14types,whichincluded1metabolismrelatedgenes,12energymetabolismrelatedgenes,5transportpathwayrelatedge
