1、基質(zhì)細(xì)胞衍生因子1在骨髓間質(zhì)干細(xì)胞歸巢到急性梗死心肌中的作用2007年6月第33卷第6期Jian~suMedJ,June007'13310.基質(zhì)細(xì)胞衍生因子1在骨髓間質(zhì)干細(xì)胞歸巢到急性梗死心肌中的作用莊瑜陳鑫石開(kāi)虎徐明【摘要】目的探討基質(zhì)細(xì)胞衍生因子1(SDF_1)在骨髓問(wèn)質(zhì)干細(xì)胞(MSCs)歸巢到急性梗死心肌中的作用.方法以免疫組化檢測(cè)SDF-1在心肌梗死1,2,4,7,14,28d后的大鼠及非心肌梗死大鼠心肌中的表達(dá)差異;于心肌梗死4d后予以SDF-1,抗SDF-1抗體進(jìn)行干預(yù),同時(shí)經(jīng)尾靜脈注射BrdU標(biāo)記的MSCs,3d后檢測(cè)
2、梗死區(qū)MSCs的歸巢量,28d后檢測(cè)大鼠心功能狀況,以及梗死區(qū)及其周?chē)难苊芏?另設(shè)心肌梗死及非心肌梗死陰性對(duì)照組.結(jié)果SDF-1在心肌梗死后第1天即升高并達(dá)到峰值,以后逐漸下降,第14天恢復(fù)至正常水平.心肌梗死組干細(xì)胞歸巢量明顯大于非心肌梗死對(duì)照組.SDF-1處理組大鼠心肌梗死區(qū)干細(xì)胞歸巢量,心功能水平及血管密度大于抗SDF-1抗體處理組及心肌梗死對(duì)照組(P<O.01).結(jié)論心肌梗死后早期梗死區(qū)SDF-1表達(dá)水平升高;SDF-1能夠促進(jìn)MSCs歸巢并改善心功能,促進(jìn)毛細(xì)血管增生可能為其改善心功能的機(jī)制之一.【關(guān)鍵詞】心肌梗
