1、腺昔預處理對糖氧剝奪損傷星形膠質(zhì)細胞的保護作用摘要:目的:觀察糖氧剝奪對星形膠質(zhì)細胞的影響及腺昔的保護作用。方法:取體外培養(yǎng)第三代或第四代星形膠質(zhì)細胞,傳代至96孔板或24孔板內(nèi),細胞貼壁并長出突起后,隨機分為三組:正常對照組(normalcontrolgro叩)、糖氧剝奪組(OGDgroup)s腺昔預處理組(ADOgroup),進行糖氧剝奪/復氧處理。觀察糖氧剝奪8h復氧糖24h后星形膠質(zhì)細胞形態(tài)、細胞活性。結果:糖氧剝奪復氧糖24h后,正常對照組星形膠質(zhì)細胞生長狀態(tài)良好;OGD模型組星形膠質(zhì)細胞胞體水腫變圓,細胞形
2、態(tài)由不規(guī)則變成梭形,突起明顯變短或消失;ADO組細胞水腫較輕。OGD模型組星形膠質(zhì)細胞活力下降,OD值明顯低于正常對照組(P<0.01);ADO組細胞活力顯著高于OGD模型組,表明ADO能明顯改善細胞活力(P<0.01)o結論:糖氧剝奪可引起星形膠質(zhì)細胞嚴重損傷,星形膠質(zhì)細胞形態(tài)、活性與缺氧缺糖呈對應變化,腺昔能減少糖氧剝奪后星形膠質(zhì)細胞形態(tài)學改變及細胞凋亡,提示腺昔對糖氧剝奪的星形膠質(zhì)細胞有一定的保護作用。關鍵詞:腺昔;星形膠質(zhì)細胞;糖氧剝奪TheprotectionofAdenosinePreconditionin

6、rmisfromirregular,andprominenceobviouslybecameshortordisappeared;theedemawaslighterinADOgroup?ActivityofastrocytecellsdecreasedinOGDgroup,andODvaluewassignificantlylowerthanthatinnormalcontrolgroup(P<0.01).CellactivitywashigherinADOgroupthanthatinOGDgroup,whichi
