2、,}L/R)損傷心肌細(xì)胞的保護(hù)作用,并探討其對ROS/PKC/p38MAPK途徑的影響。方法:分離原代乳鼠心肌細(xì)胞,建立H/R模型,細(xì)胞分為7組:正常對照組(N組)、缺復(fù)氧組(H/R組)、缺氧/復(fù)氧+PFC(終濃度20ms/L、100ms/L和200msZL)預(yù)處理組、缺氧/復(fù)氧+PFC(100ms/L)預(yù)處理+蛋白激酶C特異性阻斷劑chelerythrine(1i.Lmol/L)組及缺氧/復(fù)氧+PFC(100ms/L)預(yù)處理+p38MAPK特異性阻斷劑SB203580(10I~mol/L)組。倒置顯微鏡下觀察
3、細(xì)胞形態(tài)和自發(fā)搏動頻率,Hoechst33258染色觀察細(xì)胞凋亡率,酶標(biāo)儀測定ROS含量、SOD活性及LDH漏出量,免疫印跡法檢測PKC、p-p38MAPK和HSP70的蛋白水平。結(jié)果:H/R組細(xì)胞活力下降,搏動頻率減慢,細(xì)胞ROS含量、LDH漏出量、細(xì)胞凋亡率及p-p38MAPK的水平均較N組顯著增加(P<0.01)。經(jīng)PFC預(yù)處理后,細(xì)胞搏動頻率、SOD活性及PKC、HSP70的水平較H/R組顯著增加(P<0.05,),p-p38MAPK的水平和細(xì)胞凋亡率均減少(P<0.05),而PFC的保護(hù)作用能被che
