2、MTT、細胞計數(shù)實驗方法檢測DADS抑制NCI—H446細胞增殖;通過HE染色和AO—EB熒光染色方法,觀察DADS處理后NCI—H446細胞的形態(tài)學改變。結(jié)果:MTT結(jié)果顯示:DADS作用于NCI—H446細胞48h后,代謝MTT的能力明顯降低,顯示出較強的細胞毒性反應,Ic5o值介于20~40txg/ml之間。細胞計數(shù)結(jié)果表明:DADS作用于NCI—H446細胞后,隨DADS濃度增加NCI—H446細胞倍增時間延長。HE染色顯示:NCI—H446細胞經(jīng)DADS處理24h后,與對照組相比,細胞體積變小,胞漿豐富,細胞核變小,染色變淡。AO.EB熒光染色顯示:NCI.H446細胞經(jīng)DAD
3、S處理24h后,與對照組相比,細胞皺縮、呈圓形,胞質(zhì)黃色或橘紅色,細胞核或細胞質(zhì)內(nèi)可見致密濃染的黃綠色或橘紅色熒光,并可見橘紅色碎片且隨DADS濃度增加,隨DADS濃度增加細胞密度逐漸減少。結(jié)論:DADS能抑制體外培養(yǎng)的NCI—H446細胞增殖,作用效果與藥物濃度及作用時間相關(guān)。關(guān)鍵詞:二烯丙基二硫;小細胞肺癌;MTT中圖分類號:R734.2文獻標識碼:A文章編號:1673—6273(2014)08-1460-04ProliferationInhibitedinNCI—H440CellswithDADS:IcL1UDe-yong~,ZHANGQing-f,WANGHai-lun1,PEN
