2、取總RNA,通過RT—PCR擴(kuò)增小鼠的重鏈可變區(qū)和Linker(VH—linker)序列,以及輕鏈可變區(qū)和Linker(VK—linker)序列。經(jīng)重組PCR將兩段基因連接起來,將VH/K與pMD19一T栽體鏈接產(chǎn)物轉(zhuǎn)化E.coliDH5d,PCR鑒定并測序。[結(jié)果]VH和K鏈得到正確的擴(kuò)增,長度分別為421和689bp,VH/K連接產(chǎn)物正確,連接產(chǎn)物長度為1079bp。[結(jié)論]所采用的引物及反應(yīng)條件能夠擴(kuò)增出目的基因,成功構(gòu)建了鼠源抗ICOSL核糖體展示抗體庫,為進(jìn)行核糖體展示體外篩選抗ICOSL特異性單鏈抗體奠定基礎(chǔ)。關(guān)鍵詞ICOSL(可誘導(dǎo)共
